DExSI: A new tool for the rapid quantitation of 13C-labelled metabolites detected by GC-MS.

چکیده
Summary Stable isotope directed metabolomics is increasingly being used to measure metabolic fluxes in microbial, plant and animal cells. Incorporation of 13C/15N isotopes into a wide range of metabolites is typically determined using gas chromatography-mass spectrometry (GC/MS) or other hyphenated mass spectrometry approaches. The DExSI pipeline is an interactive graphical software package which can be used to rapidly quantitate isotopologues for a wide variety of metabolites detected by GC/MS. DExSI performs automated metabolite annotation, mass and positional isotopomer abundance determination and natural isotope abundance correction. It provides a range of output options and is suitable for high throughput analyses. Availability DExSI is available for non-commercial use from Using the password: NewDExSI2017 for Microsoft Windows 7 or higher (64-bit). Contact [email protected] or [email protected]. A Rapid and Sensitive Method for Determina-tion of Morphine in Saliva of Opium Addicts
Background: Saliva is an alternative biological matrix in drug testing. It has several advantages in comparison with other biological fluids such as blood and urine. Collection of saliva is simple, noninvasive and can be controlled under direct observation in drug abusers, or opium addicts. Methods: In the present study we report our experience with saliva collection, liquid-liquid extraction a...
متن کاملEnhanced Isotopic Ratio Outlier Analysis (IROA) Peak Detection and Identification with Ultra-High Resolution GC-Orbitrap/MS: Potential Application for Investigation of Model Organism Metabolomes
Identifying non-annotated peaks may have a significant impact on the understanding of biological systems. In silico methodologies have focused on ESI LC/MS/MS for identifying non-annotated MS peaks. In this study, we employed in silico methodology to develop an Isotopic Ratio Outlier Analysis (IROA) workflow using enhanced mass spectrometric data acquired with the ultra-high resolution GC-Orbit...
متن کاملIdentification of Volatile Organic Compounds from Trichoderma virens (6011) by GC-MS and Separation of a Bioactive Compound via Nanotechnology
Fungal volatile organic compounds (VOCs) have the potential of being used as biocontrol agents for biotechnological applications in agriculture, industry and medicine. In this research, different VOCs from secondary metabolites of biocontrol fungus Trichoderma virens (6011) KP671477 were separated using n-hexane, n-butanol and methanol solvents and identified by gas chromatography–mass spectrom...
متن کاملRapid Quantification of Major Volatile Metabolites in Fermented Food and Beverages Using Gas Chromatography-Mass Spectrometry
Here we present a method for the accurate quantification of major volatile metabolites found in different food and beverages, including ethanol, acetic acid and other aroma compounds, using gas chromatography coupled to mass spectrometry (GC-MS). The method is combined with a simple sample preparation procedure using sodium chloride and anhydrous ethyl acetate. The GC-MS analysis was accomplish...
متن کاملCertification of a new selenized yeast reference material (SELM-1) for methionine, selenomethinone and total selenium content and its use in an intercomparison exercise for quantifying these analytes.
A new selenized yeast reference material (SELM-1) produced by the Institute for National Measurement Standards, National Research Council of Canada (INMS, NRC) certified for total selenium (2,059+/-64 mg kg(-1)), methionine (Met, 5,758+/-277 mg kg(-1)) and selenomethionine (SeMet, 3,431+/-157 mg kg(-1)) content is described. The +/-value represents an expanded uncertainty with a coverage factor...
